Help with Quilt as You Go

Hi,
I'm new to the forum and also new to quilting. My first major project is a king size quilt using 12" blocks. I've made all the blocks, have the batting cut out and most of the backing. I'm starting to sandwich them together and I'm worrying about how exact they need to be. The blocks are far from perfect. Am I gonna have problems? TIA
